稳定的细胞系开发
Cellutron 提供定制的高质量稳定细胞系服务，以优化生产您感兴趣的蛋白质。基因到细胞服务包括基因合成、靶向载体构建、细胞培养、转染、稳定细胞系的药物选择和转基因表达的确认。凭借*的载体技术，Cellutron 能够在多种细胞类型中产生稳定的细胞系，包括小鼠和人类干细胞以及 iPS 细胞。
稳定细胞系-G 稳定细胞系-R
在荧光报告基因的初始转导两个月后，稳定转染的小鼠胚胎干细胞表达 GFP 或 RFP。
亲代细胞系：人和小鼠胚胎干细胞和 iPS、HEK293、CHO、BHK、NS0 细胞、H9C2 或细胞系。
稳定细胞系产生的过程和时间线：
1. 哺乳动物细胞表达载体的基因合成与构建
进行优化的基因合成并将基因克隆到哺乳动物细胞表达载体中（1 周）
2. 转基因表达分析
用表达载体转染亲本细胞系并通过 RT-PCR/Western blot/ELISA 评估转基因的表达（1 周）
3. 筛选稳定转染子
对顶级克隆进行稳定性筛选并评估最终细胞系的 10 次传代。（约 4 周）
向客户提供 2 瓶顶级稳定克隆

肌节 α 肌动蛋白的免疫细胞化学染色
心肌细胞在 60 mm 板中培养
用 1X PBS 冲洗细胞，在室温下用 PBS 中的 3.7% 甲醛固定细胞 10 分钟，然后在 70% ETOH 中 5 分钟，用 PBS 中的 0.2% Triton-X 100 孵育 5 分钟，然后用 1X PBS 洗涤细胞两次5分钟。
用 1ml 2% BSA 的 PBS 孵育细胞 30 分钟。在板中加入针对肌节 α 肌动蛋白 (Sigma, Cat#: A2172) 的原代小鼠抗体 1 小时，室温，O/N，4°C。
用 1X PBS 清洗细胞 3 次，每次 10 分钟。
与 HRP 偶联的二抗在室温下针对小鼠孵育 1 小时
用 1X PBS 清洗细胞 3 次，每次 10 分钟。
用 1 ml DAB 溶液孵育 10 分钟。用 1X PBS 清洗。
*在您需要之前准备新鲜的 DAB（Sigma，Cat#：D8001）溶液：
* 在 37 C 下解冻 DAB 原液，直到不再浑浊。将 5ul 的 DAB 原液添加到 1ml 的 1X PBS 中。加入 1ul 的 30% H2O2 并充分混合。
在显微镜下检查染色的细胞。
